A Nature Medicine systematic review and network meta-analysis of 56 randomized controlled trials (60,307 adults) compared anti-obesity medications against placebo or active comparators. Semaglutide and tirzepatide achieved the greatest weight loss, each exceeding 10% total body weight loss. Beyond weight reduction, tirzepatide showed benefits in obstructive sleep apnea and MASH remission, while semaglutide reduced major adverse cardiovascular events and knee osteoarthritis pain. The authors conclude that treatment selection should be individualized based on patients' comorbidities.
