This review article updates the 'NAD World' concept, highlighting how the enzyme eNAMPT and the molecule NMN facilitate communication between the hypothalamus, adipose tissue, and skeletal muscle to regulate aging. It posits that adipose tissue-derived eNAMPT is critical for maintaining NAD+ levels, which in turn activates SIRT1 to control longevity. This framework provides a biological basis for developing anti-aging interventions.
