Japanese researchers developed Osteo-DISCO, a tissue-clearing method enabling 3D visualization of nerves and blood vessels in murine bone. They identified specific nerve entry sites in long bones and showed that surgical ablation of these nerves impaired bone formation and regeneration. Administering CGRP, a sensory neuropeptide, suppressed the nerve-ablation-induced bone loss, and in vitro data showed CGRP directly stimulates osteoblast activity, supporting sensory nerves as regulators of osteogenesis via CGRP.
